Skin is the one goal in this library with more than one small human trial to compare. Four compounds are regularly sold for skin quality or expression lines: argireline, GHK-Cu in both its forms, and SNAP-8. This guide ranks them by what was actually tested in people, not by which cream markets itself hardest.

Argireline: the only placebo-controlled human trial here
Argireline, sold under the cosmetic name acetyl hexapeptide-8, is a six-amino-acid mimic of part of SNAP-25, a protein nerve endings use to trigger muscle contraction. The idea is a topical that softens expression lines by making that signalling slightly less efficient, a much weaker version of what botulinum toxin does by cutting the protein outright.
- Blanes-Mira and colleagues (2002), the original study, applied a 10% argireline emulsion to 10 healthy women for 30 days and reported wrinkle-depth reductions of up to 30%, a small starting study rather than proof.
- Wang and colleagues (2013) ran the first randomised, placebo-controlled trial, in 60 Chinese adults with lines around the eyes, applying argireline or placebo twice daily for four weeks. The argireline group showed improvement on both subjective assessment and skin-replica measurements; the placebo group did not.
A 2025 literature review pooled ten human studies of argireline and found some reduction in wrinkles or scars in all of them, though statistical significance varied and several tested it inside multi-ingredient products. It remains small, short-term evidence, but it is the only compound here with a controlled human trial behind it.
Delivery is the open question behind all of this. Argireline is a water-soluble peptide, and the skin barrier is built to keep such molecules out. A skin-penetration analysis summarised in a 2018 paper found that only 0.22% of an applied argireline dose permeated and stayed in the outermost skin layer, with 0.01% reaching the epidermis. How much of that reaches the small muscles under an expression line, where the peptide would need to act, is not established by the wrinkle trials above.
GHK-Cu (topical): more studies, less rigorous design
GHK-Cu is a copper-binding tripeptide with a longer list of human cosmetic studies, all summarised in a 2018 review by Pickart, who discovered the peptide, and Margolina:
- A 12-week facial cream study in 71 women with photoaging reported increased skin density and thickness and fewer, shallower fine lines.
- A 12-week eye cream study in 41 women reported GHK-Cu outperforming both placebo and a vitamin K cream on lines and wrinkles.
- A 12-week thigh-skin study reported improved collagen production in 70% of women on GHK-Cu, against 50% on vitamin C cream and 40% on retinoic acid cream.
- An 8-week nano-carrier formulation reduced wrinkle volume by 31.6% relative to a comparison peptide product.
These studies are broader in what they measured than the argireline trials, but none is described as a large, placebo-controlled trial run outside the original research circle. GHK-Cu also has extensive animal wound-healing data behind it, covered on its own page, which argireline does not.
SNAP-8: marketed as stronger, backed by less
SNAP-8 is an eight-amino-acid extension of argireline, made by the same manufacturer, aimed at the same SNAP-25 target. A peer-reviewed, placebo-controlled human trial of SNAP-8 on its own could not be found. The peer-reviewed literature that exists is analytical: a 2020 paper by Ji and colleagues developed a laboratory method for measuring SNAP-8 and applied it to microneedle-patch formulations, without testing whether the peptide reduces wrinkles. The efficacy percentages that circulate in SNAP-8 marketing trace back to manufacturer material rather than published trials.
GHK-Cu (injectable): the collagen story without a skin study
Injectable GHK-Cu is the same molecule as the topical form, sold as a research vial instead of a cream. It carries the same animal and cell data on collagen production and wound repair, but no published human trial has tested what an injection does to skin. Anyone injecting it for a skin-quality reason is relying on evidence generated from creams on skin or from wounds in animals, not from injections in people.
The US FDA has listed GHK-Cu among bulk substances for compounding that may present significant safety risks, pointing to a possible risk of immunogenicity from aggregation and peptide-related impurities in a compounded injectable form; that listing was withdrawn in April 2026, which does not amount to an approval. People with Wilson's disease or another copper-handling disorder have a specific reason for caution with any copper-containing product.
Combining these in one routine
Some formulators and DIY routines pair GHK-Cu with an expression-line peptide such as argireline or SNAP-8 on the reasoning that they target different problems, collagen and skin quality versus muscle signalling. That pairing has not been studied as a combination any more than the individual ingredients have been studied at length; each peptide's evidence above still applies on its own, not jointly.
Where retinoids fit
The GHK-Cu (topical) compound page includes a comparison with tretinoin, a prescription retinoid: "by far the strongest evidence for photoaging" of anything in that comparison, based on large drug trials rather than small cosmetic studies. None of the four peptides here has been tested head-to-head against a retinoid.
Status in the Philippines
Finished skincare products containing argireline, GHK-Cu or SNAP-8 are regulated as cosmetics by FDA Philippines, and are widely available locally. Research-vial GHK-Cu sold for injection is not a cosmetic and is not registered as a medicine. None of the four is a scheduled controlled substance under RA 9165.
