What it is
Pinealon is a synthetic peptide of just three amino acids: glutamic acid, aspartic acid and arginine, written Glu-Asp-Arg or EDR. It comes from the St Petersburg Institute of Bioregulation and Gerontology, the research institute led by Russian gerontologist Vladimir Khavinson, which also produced epitalon.
Khavinson's programme is built on the idea of "peptide bioregulators": very short peptides, originally modelled on extracts from specific animal organs, that are claimed to restore normal gene activity in the tissue they target. Pinealon is the programme's brain peptide.
Before going further, the honest summary: nearly everything published on pinealon comes from Khavinson's own group or close collaborators, much of it in Russian-language journals, and none of it has been independently replicated in a proper clinical trial.
How it works
The group's own explanation is unusual for a peptide. Most peptide drugs act on a receptor on the cell surface. Khavinson's group proposes instead that short peptides like EDR pass into cells, enter the nucleus and bind directly to DNA or to histone proteins, changing which genes are switched on.
Their evidence for this comes from three kinds of work:
- Cell studies. A 2011 paper tested pinealon on rat brain cells (cerebellar granule cells), immune cells and a nerve-like cell line under oxidative stress. It reduced the build-up of reactive oxygen species and markers of cell death in a dose-dependent way, and changed cell-cycle signalling in a way the authors said could not be explained by antioxidant action alone.
- Gene expression. A 2014 paper reported that EDR and a related peptide increased serotonin production in ageing brain cortex cells, and used computer docking to suggest the peptide matched a DNA sequence in the serotonin-making enzyme's gene.
- Modelling. The group's 2021 review brought this together into a proposed mechanism involving the MAPK/ERK pathway, cell-death proteins such as caspase-3 and p53, and antioxidant enzymes.
How a three amino acid peptide would survive digestion or reach the brain intact, and whether it really binds DNA in living animals, has not been shown by an outside lab.
What the research found
Animal and cell studies
The 2021 review from Khavinson's group lists findings in rats and mouse cells: less loss of dendritic spines in cultured neurons from a mouse model of Alzheimer's disease, higher antioxidant enzyme activity in rat brain, lower caspase-3 and better maze learning in aged rats, and more normal behaviour in rat pups exposed to high homocysteine before birth. These are small studies from the same network.
Human studies
The same review cites human reports, including oral pinealon in 72 patients living with the after-effects of traumatic brain injury, where memory improved and headaches eased, and a study reporting fewer errors on an attention test with changes in brain wave recordings. The review also states that EDR improves memory problems in elderly patients.
What those reports do not provide, at least in any accessible form, is the detail needed to judge them: clear randomisation, placebo control, blinding, prespecified outcomes and full side effect data. None has been repeated outside Russia. As evidence, they sit well below a small Western pilot trial.
Side effects reported in studies
The group's reports describe no notable side effects, but they give little safety detail and involve small numbers. There is no phase 1 safety study, no toxicology package in the public literature, and no long-term data. The absence of reported harm here says more about how the studies were reported than about the compound.

Status in the Philippines
No pinealon product is registered with FDA Philippines, so it cannot be sold here as a medicine. It is not a controlled substance under RA 9165. It turns up through research-chemical suppliers, often marketed alongside epitalon to Manila's longevity crowd. That marketing is well ahead of the evidence.
Storage
Lyophilised powder is best kept refrigerated, sealed and away from light. Once reconstituted, keep it at 2 to 8°C in the main body of the fridge rather than the door.
